You can't stick one end of a cable in the guitar and one end in the headphones and expect to hear anything. If you have a pedal that is effectively a preamp with speaker simulation (e.g. a Joyo ACTone that emulates a Vox AC30 amplifier) then you could set it up as Tx in the guitar, Rx in the pedal input...but then you will still need either another set of wireless T/R's to go from the pedal output to the headphones....or a standard guitar cable to do the same.
